Oprah's argument is with Jesus, and what he said.

Jesus claimed to be the EXCLUSIVE route to God. For he himself said, "I am the way, the truth, and the life: no man cometh unto the Father, but by me." (John 14:6)

About 1 min 30 seconds into the video Oprah clearly states "There couldn't possibly be only one way."

The second woman points out that this is not a FACT what Oprah just said (that there couldn't be only one way to God) but rather simply something that OPRAH is merely saying - in other words - her (Oprah's) opinion.

In terms of the line referring to people who have never heard the Gospel of Jesus Christ - Read Acts 10. God SENT Peter to Cornelius, a devout man who feared God, in order to tell him the gospel. People often use this line (What about people who never heard way out in the jungle) as a smokescreen. Gently remind them that THEY have heard and God will judge them based on that, then get into the story of Cornelius or the person in the jungle.
